چکیده

Background The effect of polar organic xenobiotics in the central nervous system depends on blood-brain barrier (BBB) penetration of these compounds. Newly synthesized pyridinium aldoximes (K-compounds) are promising antidotes for organophosphate intoxications. However, being highly polar, their BBB penetration is questionable. Using an in vivo model we aimed to characterize the BBB penetration of K-compounds.

A Predictive Model for Blood-Brain Barrier Penetration

Motivation. It is important to determine whether a candidate molecule is capable of penetrating the blood-brain barrier in drug discovery and development. The aim of this paper is to establish a predictive model for blood-brain barrier penetration only using two simple descriptors, molecular volume and polar surface area. Method. A dataset of 111 compounds is divided into a training set of 86 c...

Screening of Blood-brain Barrier Penetration Using the Immobilized Artificial Membrane Phosphatidylcholine Column Chromatography at the Physiological Ph

In this study, 21 structurally diverse drugs with examined central nervous system penetration were used for prediction of blood-brain barrier penetration using HPLC-UV instrument. Drugs' chromatographic capacity factors were measured by the immobilized artificial membrane presented by phosphatidylcholine column. The correlation between chromatographic capacity factor, octanol-water partition co...

A 2D-QSPR approach to predict blood-brain barrier penetration of drugs acting on the central nervous system

Drugs acting on the central nervous system (CNS) have to cross the blood-brain barrier (BBB) in order to perform their pharmacological actions. Passive BBB diffusion can be partially expressed by the blood/ brain partition coefficient (logBB). As the experimental evaluation of logBB is time and cost consuming, theoretical methods such as quantitative structure-property relationships (QSPR) can ...

Estrogen provides neuroprotection against brain edema and blood brain barrier disruption through both estrogen receptors α and β following traumatic brain injury

Objective(s):Estrogen (E2) has neuroprotective effects on blood-brain-barrier (BBB) after traumatic brain injury (TBI). In order to investigate the roles of estrogen receptors (ERs) in these effects, ER-α antagonist (MPP) and, ER-β antagonist (PHTPP), or non-selective estrogen receptors antagonist (ICI 182780) were administered. Materials and Methods: Ovariectomized rats were divided into 10 gr...

Fast calculation of molecular polar surface area as a sum of fragment-based contributions and its application to the prediction of drug transport properties.

Molecular polar surface area (PSA), i.e., surface belonging to polar atoms, is a descriptor that was shown to correlate well with passive molecular transport through membranes and, therefore, allows prediction of transport properties of drugs. The calculation of PSA, however, is rather time-consuming because of the necessity to generate a reasonable 3D molecular geometry and the calculation of ...
